Determination of Gleitzide content by fully automatic potentiometric titration

1 Preface

Gleitzide is a second-generation oral sulfonylurea hypoglycemic drug. It has a hypoglycemic effect on adult diabetic patients, can reduce platelet adhesion, decrease plasma specific viscosity, lower platelet aggregation induced by ADP, and improve microcirculation in the nail crease. It is used for adult diabetes, patients with diabetes accompanied by obesity or those with vascular lesions. The pharmacopoeia of this drug clearly stipulates the testing indicators and standards, and content is one of them.

This method involves dissolving the sample in glacial acetic acid and titrating it with perchloric acid. The endpoint of the titration is determined through dynamic titration. The data repeatability is good, the data is accurate, and the safety factor of the operation is high, which greatly reduces the risk of manual operation. It is a better choice for the content titration of this drug.

2 Instruments and reagents

2.1 Instruments

JH-T6 fully automatic potentiometric titrator, non-aqueous PH composite electrode,10mL burette

2.2 Reagents

Perchloric acid (1mol/L), glacial acetic acid.

3 Experimental Methods

3.1 Experimental Procedures

Take 0.20g of this product, accurately weigh it, add 50mL of glacial acetic acid and stir until dissolved. Titrate with perchloric acid titrant (1.0moL/L), and correct the titration result with a blank value. 1mL of perchloric acid is equivalent to 32.34㎎ gleitzite.
